Yorkshire has lifted the trophy eight times since the inaugural final in 2000, and last year beat Northumberland 53-20 at Twickenham Stadium.
They will take on Hampshire in the final four, who scored five tries at Havant RFC to beat Surrey 41-24, with Jerome Rudder responsible for a brace on his home turf.
Elsewhere, Kent remains unbeaten in the competition, after they edged Gloucester 27-25 earning a home semi-final. They will play Durham – who recorded a 31-25 victory over North Midlands.
